Overview

microRNA-569 (miR-569) is a short, non-coding RNA molecule (20–24 nucleotides) involved in post-transcriptional gene regulation, primarily by binding to the 3'-untranslated regions of target mRNAs and affecting their stability or translation. miR-569 has been identified as a tumor suppressor in pancreatic cancer, where its expression is reduced in poor prognosis cases. Overexpression of miR-569 in cell and animal models suppresses tumor cell migration, invasion, and liver metastasis, primarily by directly targeting and downregulating NUSAP1, a spindle-associated protein, and consequently inhibiting the pro-metastatic ZEB1 pathway. Low levels of miR-569 are associated with poor survival in pancreatic cancer, and it may serve as both a prognostic biomarker and a potential therapeutic target. No direct drug or small-molecule modulators of miR-569 are currently known, but altering its activity or mimicking its function could be a potential therapeutic avenue for certain cancers[1][2][4].

02

Mechanism of action

Inhibits NUSAP1 mRNA/protein translation by direct binding to its 3'-untranslated region, leading to downstream effects on ZEB1 and metastatic processes.

03

Biological functions

Post-transcriptional regulation of gene expressionNegative regulation of metastasisCell migration and invasion inhibitionCell cycle regulationApoptosis induction
04

Disease associations

Cancer (including pancreatic cancer, lung cancer, and potentially others)
05

Safety considerations

None specifically reported; as a microRNA, off-target gene regulation and tissue specificity are general concerns for any therapeutic exploitation.
06

Biomarkers

Prognostic marker for pancreatic cancer (higher expression correlates with better prognosis)
