Overview

microRNA-605 (miR-605) is a small, non-coding RNA molecule that regulates gene expression post-transcriptionally, primarily by binding the 3′ untranslated regions of target mRNAs. miR-605 is transcriptionally activated by p53 and plays a pivotal role in the p53-Mdm2 feedback loop: it suppresses Mdm2, leading to increased p53 activity and robust cellular response to stress. Functionally, miR-605 inhibits cell proliferation, migration, invasion, and promotes apoptosis in various cancer models, acting as a tumor suppressor. It has shown altered expression in multiple cancers and its genetic variants or expression levels have been linked to cancer susceptibility and prognosis. No approved drugs directly target miR-605, but it is being considered as a therapeutic tool and biomarker in oncology research.

Mechanism of action

Acts as a tumor suppressor or inhibitor of oncogenicity by repressing its mRNA/protein targets (notably Mdm2, FOXP1, INPP4B, SOX9). Positive regulation of p53 by relieving p53 inhibition through Mdm2 repression.
