MicroRNA-616 (miR-616) is a small non-coding RNA molecule that functions as a critical post-transcriptional regulator of gene expression by binding to the 3' untranslated regions (UTRs) of target messenger RNAs [PMID: 30106411]. It is primarily recognized as an oncomiR, frequently overexpressed in various human malignancies such as non-small cell lung cancer, hepatocellular carcinoma, and prostate cancer [PMID: 29568354, PMID: 21224345]. miR-616 promotes oncogenic phenotypes, including increased cell proliferation, migration, and epithelial-mesenchymal transition (EMT), by directly targeting and silencing tumor suppressor genes like PTEN, SOX7, and TFPI-2 [PMID: 28654468, PMID: 21224345]. In addition to its role in cancer, miR-616 has been implicated in inflammatory responses and the regulation of the Wnt/beta-catenin and PI3K/Akt signaling pathways [PMID: 38016544, PMID: 28654468]. While no clinical-stage drugs are currently approved for this target, experimental strategies using antisense oligonucleotides (antagomirs) to inhibit miR-616 are being explored as potential therapeutic interventions to restore tumor suppressor activity [PMID: 32445145].
