Overview

MicroRNA 630 is a small non-coding RNA belonging to the microRNA family, encoded on chromosome 15q24.1. miR-630 acts as a post-transcriptional regulator by binding target mRNA 3'-UTRs, leading to their degradation or translational repression. It exhibits complex, context-dependent functions: it can inhibit cell proliferation and induce cell cycle arrest by targeting CDC7 kinase, or modulate apoptotic balance under genotoxic stress by regulating pro- and anti-apoptotic genes such as PARP3, DDIT4, EP300, p53, and BCL2. Its expression levels correlate with tumor progression, metastatic potential, and chemoresistance in several cancers—being either upregulated or downregulated depending on tumor type and biological context. Therapeutic modulation of miR-630—for example, via antisense oligonucleotides, mimics, or inhibitors—is under investigation as a potential approach in cancer therapy, mainly aiming to improve sensitivity to chemotherapeutics (e.g., cisplatin) and suppress metastatic behavior. However, given its dual (tumor-suppressive and oncogenic) roles depending on tissue and disease state, careful assessment is required for any clinical application.

Mechanism of action

Down-regulation or inhibition of miR-630 may suppress cell proliferation, migration, and invasion, and enhance apoptosis, contributing to anti-tumor effects. miR-630 modulates the response to DNA-damaging agents (e.g., cisplatin) via suppression of CDC7 kinase and multiple apoptotic regulators. miR-630 regulates EMT and metastatic potential by targeting transcription factors such as Slug in hepatocellular carcinoma. In breast cancer, miR-630 targets metadherin (MTDH), impacting the AKT, NFκB, and Wnt/β-catenin signaling pathways.
