MicroRNA 639 (MIR639) is a small, non-coding RNA molecule that post-transcriptionally regulates gene expression by binding to target messenger RNAs, leading to their repression or degradation. It is processed from a precursor stem-loop structure and functions through incorporation into the RNA-induced silencing complex (RISC). MIR639 plays a key role in carcinogenesis, with frequent downregulation observed in several tumors, including colorectal, liver, breast, ovarian, bladder, and tongue cancers. In colorectal cancer, miR-639 is notably downregulated and acts as a tumor suppressor by affecting major signaling pathways such as MAPK, Wnt, PI3K-Akt, p53, and TGF-β. Though no clinical drugs directly target miR-639, its altered expression is investigated for diagnostic and prognostic applications, and its regulatory complexity poses challenges for therapeutic targeting.
