microRNA 641 is a small, noncoding RNA (~18–25 nucleotides) that suppresses gene expression by targeting mRNA transcripts for degradation or translational inhibition. It acts as a tumor suppressor in multiple cancer types, where its expression is commonly downregulated. miR-641 inhibits cell proliferation and migration, promotes apoptosis, and modulates major signaling pathways such as PI3K/AKT (targeting NUCKS1), p53 (targeting MDM2), and EMT via ZEB1. Its tumor-suppressive effects make it an attractive candidate for diagnostic, prognostic, and therapeutic applications in oncology. Low miR-641 levels correlate with aggressive tumor features, poor prognosis, and therapy resistance. No approved drugs currently target miR-641 directly, but experimental use of miRNA mimics has demonstrated therapeutic benefit in preclinical cancer models.
In cancer, miR-641 mimics can be used to restore tumor suppressor activity by targeting oncogenes such as NUCKS1 and ZEB1. Downregulation of MDM2 leading to restoration of p53 activity. Potentially increases sensitivity to drugs like cisplatin by promoting apoptosis.
