Overview

MicroRNA 645 is a non-coding microRNA located at chromosome 20q13.13 and is frequently upregulated in colon cancer due to DNA copy number gains. Its expression promotes colorectal cancer cell proliferation, cell cycle progression, and resistance to apoptosis, largely by regulating (and partially suppressing) tumor suppressor proteins such as SOX30. Inhibition of miR-645 enhances sensitivity to chemotherapeutics (e.g., 5-fluorouracil, cisplatin), and suppresses tumor growth in vivo. Beyond colon cancer, elevated miR-645 is also associated with negative prognosis in other malignancies, including gastric cancer and head and neck squamous cell carcinoma. miR-645 acts as an oncogenic regulator and could be developed as a biomarker for early diagnosis or as a therapeutic target in cancer.
