MiR-653 displays context-dependent functions across different cancer types: it acts as an oncogene in several carcinomas (including colorectal, gastric, and prostate cancers), promoting cell proliferation, invasion, migration, EMT, and suppressing apoptosis, particularly by targeting tumor suppressors like KLF6 and SOCS6 and modulating key signaling networks such as JAK/STAT. Conversely, in other settings such as breast, liver, and lung cancers, miR-653 has been reported to inhibit tumor progression by targeting molecules like MAPK6. Its expression levels are often dysregulated in tumors and can serve as tissue or circulating prognostic biomarkers. Despite promising roles in both carcinogenesis and tumor suppression, the dual and context-dependent functions of miR-653 necessitate further research before it can be safely targeted therapeutically[1][2][3][4].
