Overview

MicroRNA 655 (miR-655) is a short, non-coding regulatory RNA (microRNA) involved in the post-transcriptional regulation of gene expression. By binding to the 3'-untranslated regions of specific mRNAs, miR-655 suppresses their translation or facilitates degradation, impacting multiple cellular processes. MiR-655 acts as a tumor suppressor in various cancers—including hepatocellular carcinoma, esophageal squamous cell carcinoma, and oral squamous cell carcinoma—by inhibiting cell proliferation, migration, and invasion and suppressing the epithelial-mesenchymal transition (EMT). Its direct molecular targets include genes such as ADAM10, ZEB1, TGFBR2, PTTG1, and metadherin, which are associated with tumor progression and signaling pathways like β-catenin and TGF-β. Downregulation of miR-655 is often correlated with increased malignancy and poorer prognosis, and restoration of its expression suppresses metastasis and invasiveness, making it a promising biomarker and candidate for therapeutic intervention in cancer[1][2][3][4][5].
