MicroRNA 665 (miR-665 or MIR665) is a short (20-24 nucleotide) non-coding RNA molecule encoded at chromosome 14q32.2 that regulates gene expression post-transcriptionally. It operates by binding to complementary mRNA sequences, resulting in either mRNA degradation or translational repression through the RNA-induced silencing complex (RISC). miR-665 has diverse roles in cancer, functioning as a tumor suppressor or oncogene via regulation of cell proliferation, apoptosis, migration, and drug resistance. It targets a range of genes (e.g., MYC, LIM and SH3 domain protein 1, HOXA10, ABCC2, TGFBR1, EXT1, MAPK1) involved in oncogenic and drug-resistance pathways. miR-665 is being evaluated as a non-invasive biomarker for early cancer detection and prognosis, although its variable function across different cancer types necessitates careful validation. Other roles include regulation of inflammation and cell signaling in other diseases.
Drugs that interact with the biological pathways regulated by miR-665 (e.g. inhibition of ERK1/2, modulation of Wnt/β-catenin or drug efflux transporters) could have altered efficacy or resistance profiles due to miR-665’s regulation of target genes. Mechanistic action involves post-transcriptional gene silencing or repression via incorporation into the RNA-induced silencing complex (RISC).
