Overview

MicroRNA 6744 (miR-6744) is a small non-coding RNA molecule that regulates gene expression post-transcriptionally by binding to target mRNAs and reducing their translation or promoting degradation. MiR-6744-5p plays a key tumor suppressive role in breast cancer cell lines by promoting anoikis, a form of apoptosis induced by loss of cell attachment to the extracellular matrix, thereby acting as a barrier to metastasis. Experimental studies show that miR-6744-5p is downregulated in anoikis-resistant breast cancer sublines, and overexpression restores sensitivity to anoikis while decreasing migration and invasiveness. The N-acetyltransferase 1 (NAT1) enzyme was identified as a direct downstream target of miR-6744-5p, linking its regulatory function to key processes in cell survival, EMT, and metastatic progression. Expression levels of miR-6744-5p, as well as its impact on E-cadherin, highlight its importance as a potential therapeutic target and biomarker in breast cancer.

Mechanism of action

miR-6744-5p functions by downregulating target mRNA, specifically the N-acetyltransferase 1 (NAT1) enzyme, resulting in increased sensitivity to anoikis and decreased migration/invasion in breast cancer cells

03

Biological functions

Post-transcriptional gene regulationPromotion of anoikis (cell death upon loss of adhesion)Regulation of epithelial-mesenchymal transition (EMT) via E-cadherinSuppression of cell migration and invasion in cancer cells
04

Disease associations

Cancer (specifically breast cancer metastasis and progression)Other (potential implications in additional epithelial cancers via EMT regulation)
05

Biomarkers

miR-6744-5p expression can serve as a biomarker for anoikis sensitivity, EMT status, and possibly metastatic potential in breast cancer
