microRNA 6745 (miR-6745)

Target
miR-6745
Molecular classification
microRNA, Non-coding RNA, Regulatory RNA
01

Overview

miR-6745 is a human microRNA that acts as a post-transcriptional regulator by binding to the 3'-untranslated region (3'-UTR) of target mRNAs, notably those encoding tissue inhibitor of metalloproteinases 1 (TIMP1). In gastric cancer, miR-6745 expression is decreased, which contributes to tumor progression. Experimental restoration of miR-6745 in cancer cell and mouse models suppresses tumor growth and metastasis by downregulating TIMP1 and inhibiting the canonical Wnt/β-catenin signaling pathway, suggesting its potential utility as a therapeutic target or biomarker for patient prognosis in gastric cancer. Its roles in other tissues and disease settings are not yet well characterized.

02

Mechanism of action

Suppresses oncogenesis by binding to the 3'-UTR of TIMP1 mRNA, leading to its downregulation and subsequent inhibition of Wnt/β-catenin signaling

03

Biological functions

Post-transcriptional regulation of gene expressionTumor suppressionInhibition of cell proliferationInhibition of cell migration and invasionModulation of Wnt/β-catenin signaling pathway
04

Disease associations

Cancer (specifically gastric cancer where it acts as a tumor suppressor)Potential roles in other cancers due to its regulatory nature, though currently direct evidence is limited to gastric cancer
05

Safety considerations

No notable safety concerns or therapeutic challenges directly attributed to manipulation of miR-6745 have been reported in the context of published *in vitro* or *in vivo* models; safety concerns would be expected for any RNA-based therapy but data are lacking.
06

Biomarkers

Low miR-6745 expression in gastric cancer tissue (potential prognostic biomarker for poor outcomes)Negative correlation with TIMP1 protein levels in gastric cancer tissue
