MicroRNA 718 (miR-718 or MIR718) is a short, non-coding RNA molecule involved in post-transcriptional regulation of gene expression by binding to complementary sequences within target mRNAs, thereby causing their degradation or translational inhibition. It is part of the miRNA family and is processed from a stem-loop precursor to a mature miRNA as part of the RNA-induced silencing complex (RISC)[3]. miR-718 specifically regulates the intensity and duration of innate immune responses by downregulating phosphatase and tensin homolog (PTEN), thereby modulating PI3K/Akt signaling, and subsequently repressing proinflammatory cytokine production and activity of the NF-κB pathway[1]. It has a tumor-suppressive effect in non-small cell lung cancer through direct targeting of cyclin B1 (CCNB1) mRNA, resulting in reduced cell proliferation and increased apoptosis[2]. Decreased miR-718 expression is associated with greater bacterial burden in infection and altered cancer progression. MIR718 overlaps with the 5′ UTR of the IRAK1 gene, enabling tight regulatory feedback of inflammatory signaling. MIR718 is not a classical drug target such as a receptor, enzyme, or transporter, but rather acts at the RNA level to orchestrate cellular pathways relevant to disease and homeostasis[1][2][3].
