Overview

MicroRNA-769 (miR-769) is a regulatory non-coding RNA molecule classified as a microRNA that modulates gene expression at the post-transcriptional level, primarily by binding to complementary sequences in the 3'-UTR of target mRNAs, thereby inhibiting their translation or promoting degradation. It exists as two mature forms (miR-769-3p and miR-769-5p), each potentially targeting different transcripts. miR-769-3p and miR-769-5p have been identified as having tumor-suppressive functions in multiple human cancers, including glioma, colorectal cancer, and oral squamous cell carcinoma, by inhibiting genes such as ZEB2, NDRG1, MAPK1, and NUSAP1. miR-769 contributes to the regulation of cell proliferation, migration, invasion, cell cycle progression, and apoptotic pathways. Its expression levels are associated with prognosis, chemoresistance, and disease subtype in several cancers, making it both a potential therapeutic target and a biomarker for diagnosis and treatment response. However, therapeutic targeting of miR-769 presents challenges due to potential off-target effects and the pleiotropic roles of miRNAs in gene networks[1][2][3][4][5][6][7].

Mechanism of action

Inhibition of mRNA translation or promotion of mRNA degradation of specific oncogenes and pathway components, including ZEB2 and NDRG1, among others. Suppression of Wnt/β-catenin signaling pathway via targeting ZEB2 in glioma. Inhibition of MAPK1 (notably by miR-769-5p in colorectal cancer cells). Inhibition of NUSAP1 (tumor suppressor mechanism in bladder cancer).
