Overview

MicroRNA-874 (miR-874) is a small, non-coding RNA molecule located on chromosome 5q31.2 that functions primarily as a tumor suppressor by regulating the post-transcriptional silencing of multiple target genes across diverse biological pathways[4][7]. miR-874 is downregulated in numerous cancers and other diseases, and its decreased expression is linked to enhanced cell proliferation, reduced apoptosis, metastasis, and chemotherapy resistance. miR-874 acts through direct targeting of genes such as XIAP, PMVK, PPP1CA, and through involvement in ceRNA networks. It plays a central role in modulating major signaling pathways (e.g., Wnt/β-catenin, PI3K/AKT, Hippo), and its restoration suppresses tumor growth and enhances chemosensitivity in model systems[2][4][7]. Due to its downregulation and tumor suppressor function, miR-874 is considered both a biomarker and a potential therapeutic target in cancer diagnosis, prognosis, and treatment resistance.
