Overview

microRNA-874-5p (miR-874-5p) is a small, non-coding, single-stranded RNA of about 22 nucleotides transcribed from the human genome. It functions by binding to complementary sequences in the 3′ untranslated regions (3′ UTR) of target mRNAs, leading to inhibition of gene expression at the post-transcriptional level. miR-874-5p and its partner miR-874-3p are typically transcribed as a precursor hairpin, then processed and sorted into 5p and 3p mature forms. miR-874-5p is frequently downregulated in multiple tumor types and is considered a tumor suppressor, as its restoration inhibits proliferation, migration, and invasion, and induces apoptosis in cancer cells. It participates in complex regulatory networks (including ceRNA axes), affects vital signaling pathways (such as Wnt/β-catenin, STAT3/VEGF-A, PI3K/AKT), and is involved in non-cancerous conditions like inflammation, ischemia, and neuronal injury. Its dysregulation is associated with both malignancy and other disease states, making it an important target for biomarker development as well as a potential candidate for therapeutic RNA-based interventions.
