Overview

MicroRNA 9-2 (MIR9-2) is one of three genomic loci in humans (along with MIR9-1 and MIR9-3) that produce an identical mature microRNA: miR-9. MiR-9 is a highly conserved, brain-enriched, small non-coding RNA of ~22 nucleotides that post-transcriptionally regulates gene expression by targeting messenger RNA transcripts. Through its regulatory function, miR-9-2 plays essential roles in neurogenesis, affecting the proliferation, migration, and differentiation of neural progenitor cells, as well as in controlling cell proliferation and apoptosis in other cell types. In cancer, particularly acute myeloid leukemia with MLL rearrangements, miR-9-2 is specifically upregulated and promotes cell survival, proliferation, and transformation, thus behaving as an oncomiR and potential therapeutic target. Its dysregulation is also implicated in various solid tumors and neurodegenerative diseases. Experimental therapeutics, including antagomiRs that inhibit miR-9-2, show potential in disease models but carry risks due to miR-9-2’s critical roles in neural development and homeostasis.
