The microRNA (miRNA) biogenesis machinery is a multi-component enzymatic pathway responsible for the production of mature miRNAs from primary transcripts [Ha and Kim, 2014, Nature Reviews Molecular Cell Biology]. This process begins in the nucleus, where the Microprocessor complex, consisting of the RNase III enzyme Drosha and the RNA-binding protein DGCR8, cleaves primary miRNAs (pri-miRNAs) into precursor miRNAs (pre-miRNAs) [Gebert and MacRae, 2019, Nature Reviews Molecular Cell Biology]. These precursors are exported to the cytoplasm by Exportin-5 and further processed by the RNase III enzyme Dicer and its cofactor TRBP into mature miRNA duplexes [Lin and Gregory, 2015, Nature Reviews Cancer]. The mature miRNA is then loaded into the RNA-induced silencing complex (RISC), where it guides Argonaute proteins to suppress target gene expression through mRNA degradation or translational inhibition [Ha and Kim, 2014, Nature Reviews Molecular Cell Biology]. Dysregulation of this machinery is a hallmark of many diseases, particularly cancer, where global miRNA downregulation often promotes tumor progression [Lin and Gregory, 2015, Nature Reviews Cancer]. Therapeutic interventions include small molecules like enoxacin, which enhances miRNA processing to restore tumor-suppressive functions, and aminoglycosides that can inhibit specific processing steps [Melo et al., 2011, Nature Medicine; Wang et al., 2008, Biochemistry]. However, targeting this machinery presents significant challenges, primarily due to the potential for widespread off-target effects and global disruption of cellular gene regulation [Gebert and MacRae, 2019, Nature Reviews Molecular Cell Biology].
