MicroRNA let-7i target messenger RNAs (mRNAs) are a collection of transcripts regulated by the let-7i microRNA, a key member of the let-7 family known for its role in development and tumor suppression (Roush & Slack, 2008; PMID: 18424138). These targets include prominent oncogenes such as HMGA2, KRAS, and MYC, as well as immune-related genes like TLR4 (Chen et al., 2007; PMID: 17635982). By binding to complementary sequences in the 3' untranslated regions (UTRs) of these mRNAs, let-7i facilitates their degradation or inhibits their translation, thereby modulating various cellular pathways including cell cycle progression and inflammatory signaling. In many cancers, let-7i is downregulated, leading to the overexpression of its target mRNAs and promoting malignancy and metastasis (Boyerinas et al., 2010; PMID: 20473324). Therapeutic approaches targeting this axis often involve the use of miRNA mimics to restore the silencing of these transcripts or antagomirs to block let-7i activity in specific contexts like chronic inflammation or fibrosis (Long & Slack, 2021; PMID: 33510477).
