Microtubule-actin cross-linking factor 1 (MACF1), also known as ACF7, is a massive cytoskeletal linker protein and a member of the spectraplakin family[1][2][5]. MACF1 integrates the actin microfilament, microtubule, and, in some isoforms, intermediate filament networks, acting as a key organizer of cytoskeletal dynamics[2][3]. The N-terminal actin-binding domain and C-terminal microtubule interaction modules allow the protein to physically connect these cytoskeletal elements, with further regulation via post-translational modifications such as phosphorylation[1][2]. MACF1 is essential for embryonic development and regulates critical cellular processes including migration, cell polarity, proliferation, and maintenance of tissue integrity[2][3][4]. Disruption of MACF1 function has been implicated in neurological disorders, certain cancers, and defects in tissue organization and repair[2][4]. The protein is not currently considered a direct therapeutic target and is not known to be modulated by clinically approved drugs[2][5].
