Microtubule-associated protein 1B (MAP1B) is a critical cytoskeletal protein encoded by the MAP1B gene, predominantly expressed in the developing and adult nervous system[1][2][4][5][8]. It is a large polyprotein that undergoes proteolytic processing into heavy and light chains, both of which associate with microtubules. MAP1B facilitates microtubule assembly, stability, and cross-talk with the actin cytoskeleton, processes fundamental to neurogenesis, axonal guidance, elongation, and synaptic plasticity[1][2][4][6][8]. Recent research shows MAP1B also modulates mitochondrial localization in neurons, regulates membrane trafficking, and influences autophagy by interacting with other proteins such as syntaxin 17[3]. Loss or mutation of MAP1B results in significant neurodevelopmental defects, underlining its essential role in nerve cell architecture and signaling[1][4][6]. The protein is not currently an established drug target, and no therapies directly modulate MAP1B in clinical practice.
