Overview

Microtubule-associated protein 6 (MAP6), also known as stable tubule-only polypeptide (STOP), is a calmodulin-binding protein encoded by the MAP6 gene in humans[1][5][9]. MAP6 is best known for its role in stabilizing microtubules against conditions that typically promote depolymerization, such as cold and exposure to nocodazole[1][2][3][4][9]. It contains distinctive Mn and Mc motifs, which are critical for its microtubule stabilization properties[1][2][3][4]. MAP6 is highly expressed in the nervous system, primarily in neurons, and is involved in regulating the structure and function of the neuronal cytoskeleton, including dendritic spine formation and maintenance[3][4]. MAP6 interacts with synaptic components and is essential for proper synaptic plasticity, learning, and memory[4]. Although it has been implicated in processes relevant to neurodevelopmental and neurodegenerative diseases, MAP6 itself is not a direct therapeutic target (e.g., it is not a receptor, enzyme, or transporter). Its deficiency in mouse models leads to profound deficits in synaptic function and behavior, highlighting its physiological importance[4].
