Overview

Microtubule-associated protein tau (MAPT) is a neuronal protein that stabilizes microtubules and regulates axonal transport. Phosphorylation regulates tau's function; hyperphosphorylation leads to detachment from microtubules, aggregation into neurofibrillary tangles, and neurodegeneration in Alzheimer's disease and other tauopathies. Key phosphorylation sites include Ser199/Ser202/Thr205, Thr212, Thr231/Ser235, Ser262/Ser356, Ser396/Ser404, and Ser422. Kinases such as MARKs phosphorylate tau. Mutations can promote increased phosphorylation rates.
