Microtubule-associated tyrosine carboxypeptidase 1 (MATCAP1) is an enzyme that removes the C-terminal tyrosine residue from α-tubulin, a key component of microtubules, thus regulating microtubule dynamics and cellular function[2][6][8]. This post-translational modification, known as detyrosination, affects microtubule stability and has essential roles in the nervous system, including brain development and modulation of astrocyte structural plasticity[1][2][4]. MATCAP1 activity is primarily cytosolic and associated with the cytoskeleton, and it may contribute to reactive astrogliosis if dysregulated[1][4]. No specific drugs are currently known to interact directly with MATCAP1, and there are no established biomarkers or approved therapies targeting this enzyme in clinical practice. However, due to its critical role in cytoskeletal regulation, inhibition or dysregulation could have significant neurological consequences[1][5].
