Microtubule beta-tubulin is a key component of microtubules, which are dynamic cytoskeletal polymers essential for cell division, intracellular transport, and cell shape. Taxanes, such as paclitaxel and docetaxel, bind to β-tubulin within assembled microtubules, stabilizing them and preventing depolymerization. This disruption of microtubule dynamics leads to mitotic arrest and apoptosis, making β-tubulin a crucial target for cancer therapy. However, taxane binding can also induce structural distortions that contribute to peripheral neurotoxicity. Drug resistance mechanisms, like P-glycoprotein efflux, also pose therapeutic challenges.
