Overview

The microtubule-organizing center (MTOC) is a specialized cellular structure found in eukaryotic cells that serves as the primary site for microtubule nucleation and organization. In animal cells, the principal MTOC is the centrosome, which consists of a pair of centrioles surrounded by pericentriolar material (PCM) rich in gamma-tubulin ring complexes. The MTOC plays a critical role in establishing the cell's internal architecture, facilitating intracellular transport, and directing the assembly of the mitotic spindle during cell division. Dysregulation of the MTOC, particularly centrosome amplification, is a hallmark of many cancers, leading to genomic instability and aneuploidy. Consequently, components of the MTOC, such as Polo-like kinase 4 (PLK4) and Aurora kinases, have emerged as therapeutic targets to selectively kill cancer cells that rely on supernumerary centrosomes. Drugs targeting these components aim to disrupt centriole duplication or spindle pole integrity, leading to mitotic arrest and apoptosis.
