MINDY lysine 48 deubiquitinase 3 (MINDY3) is an enzyme in humans that belongs to the MINDY (MIU-containing novel DUB family) class of deubiquitinase enzymes, which consists of four members. MINDY3 specifically targets and removes ubiquitin from lysine 48-linked polyubiquitin chains, which signals for proteasomal degradation. By reversing ubiquitination, MINDY3 regulates protein turnover and homeostasis and may play a role in apoptosis and tumor suppression. Like other deubiquitinases (DUBs), MINDY3 functions as a cysteine protease, acting as an exo-DUB (removing ubiquitin units from the distal end of polyubiquitin chains). Deregulation can affect cellular signaling and has implications in cancer, as MINDY3 has been identified as a tumor suppressor. No specific therapeutics are currently approved that directly target MINDY3, but it is recognized as a potential therapeutic target in diseases involving ubiquitin pathway disruption.
