Overview

Mineral ions bound by casein and whey proteins refer to the physiological and chemical complexes formed between milk proteins and essential divalent cations like calcium, magnesium, and zinc. Caseins, particularly alpha-s1 and beta-casein, are phosphoproteins that form large colloidal aggregates called micelles, which sequester amorphous calcium phosphate to prevent its precipitation (Holt et al., 2013, PubMed: 23465093). Whey proteins, such as alpha-lactalbumin, function as metalloproteins with high-affinity binding sites for calcium, which is necessary for their structural stability (UniProt: P00709). These protein-mineral interactions are critical for increasing the bioavailability of minerals in the neonatal diet and are exploited in therapeutic contexts. For example, Casein Phosphopeptide-Amorphous Calcium Phosphate (CPP-ACP) complexes are used to promote dental enamel remineralization and prevent caries (Reynolds, 2008, PMID: 19091344). While these complexes are not traditional drug targets for small molecule inhibition or activation, they serve as vital delivery systems for micronutrients and are used to manage conditions like osteoporosis and dental erosion. The binding of minerals to these proteins also protects the minerals from forming insoluble salts with phytates or oxalates in the gut, representing a key mechanism for mineral homeostasis and delivery in mammalian biology.
