The mineralized tooth matrix is a complex, highly organized extracellular structure that constitutes the hard tissues of the tooth, including enamel, dentin, and cementum (StatPearls, Physiology, Tooth). It is primarily composed of inorganic hydroxyapatite crystals embedded within or supported by an organic framework, which is protein-rich in dentin (mostly Type I collagen) and nearly absent in mature enamel (Journal of Dental Research, Dentin Matrix Proteins). Biologically, this matrix provides the mechanical strength necessary for mastication while protecting the underlying vital pulp from thermal, chemical, and bacterial insults. In disease states such as dental caries, the matrix undergoes demineralization due to organic acids produced by plaque bacteria, leading to structural failure (NIH, Dental Caries). Therapeutic interventions often target this matrix to promote remineralization, using agents like fluoride which substitute hydroxyl groups in hydroxyapatite to form more acid-resistant fluorapatite (CDC, Fluoride Products). Additionally, the matrix serves as a substrate for adhesive restorative materials and desensitizing agents that occlude exposed dentinal tubules.
