Overview

Minichromosome maintenance complex-binding protein (MCMBP) is a highly conserved, non-enzymatic chaperone essential for the assembly of the MCM2–7 hexamer, the replicative DNA helicase central to eukaryotic DNA replication. MCMBP interacts with MCM3 and to a lesser extent with other MCM subunits, aiding the stable incorporation of newly synthesized MCM3 and MCM5 into the hexamer. This activity is vital for maintaining a robust pool of licensing-competent MCM2–7, supporting proper cell cycle progression and genomic stability. Loss or depletion of MCMBP results in decreased MCM2–7 levels, impaired replication licensing, cell cycle arrest (in p53-competent cells), and cell death (in p53-deficient cells), indicating its potential as a therapeutic target, especially for p53-deficient cancers. There are currently no drugs that directly target MCMBP, but its role in DNA replication and maintenance of cellular proliferation marks it as a protein of interest in anti-cancer strategies.
