Overview

Minichromosome maintenance complex component 3 associated protein (MCM3AP) is an acetyltransferase that binds to and acetylates MCM3, a key DNA replication licensing factor[1][3][4]. MCM3AP inhibits the initiation of DNA replication by modifying MCM3, ensuring that DNA synthesis occurs only once per cell cycle. It interacts with components critical for chromatin structure and nuclear localization. While the MCM3AP gene is entirely contained within the GANP (germinal-center associated nuclear protein) locus, the two proteins are independently transcribed, localized to different subcellular compartments, and function distinctly. Dysfunction or allelic variation in MCM3AP is linked to neuropathies and intellectual disability. Despite its molecular role, MCM3AP is not a known target of approved drugs but is studied as a regulator of DNA replication, chromatin biology, and immune cell function[1][3][4].
