Minichromosome maintenance complex component 8 (MCM8) is an ATP-dependent DNA helicase that forms a functional complex with MCM9, known as the MCM8/9 complex. This complex plays an essential role in the initiation of DNA replication, progression and stabilization of replication forks, and repair of DNA double-strand breaks via homologous recombination. MCM8 is critical for genome integrity; its malfunction or overexpression is implicated in cancer development, tumor progression (such as in osteosarcoma), and infertility. MCM8 serves as a molecular motor regulating DNA metabolism in both normal and pathological contexts, with ongoing research into its potential as a therapeutic target and biomarker for cancer and genetic disorders[1][2][4][8].
Inhibition of helicase activity may block DNA replication and repair, leading to reduced cell proliferation and increased apoptosis in tumor cells[4].
