Overview

Minichromosome maintenance protein 10 (MCM10) is a highly conserved nuclear protein essential for the initiation of DNA replication in eukaryotic cells[2][3]. It functions as a DNA replication initiation factor by linking the MCM2–7 helicase complex to DNA polymerase and other replication proteins, facilitating replication origin firing during the S phase of the cell cycle[1][2][3]. Structurally, human MCM10 forms a hexameric ring that serves as a protein–protein interaction hub within the replication machinery[2]. MCM10 is required for proper cell cycle progression and genomic stability; its overexpression is strongly associated with increased cell proliferation and is implicated in the progression and aggressiveness of several cancers[1]. Its roles in disease and cell biology make it a candidate biomarker and a potential (though not currently established) therapeutic target, especially in oncology[1][3]. No approved drugs currently target MCM10 directly, but its centrality to replication makes it of significant research interest.
