miR-101-2 precursor microRNA (miR-101-2)

Target
miR-101-2
Molecular classification
microRNA, non-coding RNA, gene regulator
01

Overview

miR-101-2 precursor microRNA is a small, non-coding RNA located on chromosome 9p24.1; together with miR-101-1, it encodes miR-101, a highly conserved tumor suppressor implicated in multiple cancers and other diseases. It regulates gene expression post-transcriptionally by directing RISC-mediated mRNA silencing or degradation. miR-101-2 is involved in apoptosis, cell cycle, migration, and autophagy, and serves as a biomarker for cancer prognosis and patient stratification. Both its dysregulation and potential for therapeutic targeting are active areas of research.

Mechanism of action

Gene silencing (via RISC complex), mRNA degradation, and translational inhibition. Therapeutics would act by restoring or inhibiting miR-101 function, affecting its downstream targets such as EZH2, SOX9, and RAB5A.

Biological functions

Gene expression regulation (post-transcriptional silencing)ApoptosisCell proliferationCell migration/invasionAutophagy inhibitionTumor suppression
04

Disease associations

Cancer (as a tumor suppressor; down-regulated in various cancers)Fibrosis (pulmonary, hepatic)Cardiovascular diseaseNeurodegenerative disease (potential biomarker in Parkinson’s disease; see downregulation in patients)Other (angiogenesis, metastasis)
05

Safety considerations

Off-target gene regulationPotential for unwanted activation/suppression of pathways important for normal tissue functionDelivery barriers for miRNA-based therapies
06

Biomarkers

miR-101 (including the miR-101-2 precursor) is a biomarker for cancer diagnosis, prognosis, and therapeutic response. Its downregulation is correlated with tumor size, grade, and poor survival in cancers such as gastric, lung, breast, oral squamous cell carcinoma
