miR-3140-regulated oncogenic mRNAs refer to a group of messenger RNAs that are post-transcriptionally regulated by the microRNA miR-3140, particularly its mature form miR-3140-3p. This microRNA typically functions as a tumor suppressor by binding to the 3' untranslated regions (UTRs) or coding sequences of specific oncogenes, leading to their degradation or the inhibition of their translation (Yuan et al., 2016, PubMed: 26823710). Key validated targets include MAPK1 (also known as ERK2) and NRAS, which are central components of the mitogen-activated protein kinase (MAPK) signaling pathway (Uchino et al., 2013, PubMed: 23904689). In many cancers, miR-3140 is downregulated, leading to the overexpression of these oncogenic mRNAs and subsequent uncontrolled cell proliferation and survival. Therapeutic approaches focus on the use of miR-3140 mimics to restore normal regulation and suppress the expression of these oncogenic drivers. However, the complexity of miRNA targeting, including the potential to target genes like BRCA1, presents a challenge for clinical development due to potential context-dependent effects (Uchino et al., 2015, PubMed: 25672304). The interaction between miR-3140 and its targets remains a significant area of research for developing novel RNA-based therapeutics in oncology.
