Overview

The MIR142 host gene (MIR142HG) encodes a long non-coding RNA that is the primary transcript for microRNA-142, highly specific to the hematopoietic lineage. Mature miR-142 (produced from MIR142HG) is a post-transcriptional gene expression regulator that controls critical processes in immune cell differentiation, function, and homeostasis, as well as hemostasis. The gene is highly conserved and both the 3p and 5p strands of miR-142 are functionally important. MIR142HG mutations, especially those affecting the seed regions of miR-142-3p, are implicated in the pathogenesis of multiple hematological malignancies, including leukemia and lymphoma, and impact immune regulation and hematopoietic lineage development. While MIR142HG itself is not a traditional drug target, the mature miR-142 produced from it is emerging as a potential target for oligonucleotide therapeutics in oncology and immune modulation[10][3][2][4][12][7][5].
