Overview

MIR31 host gene (MIR31HG) is a long non-coding RNA gene located on chromosome 9p21.3, consisting of four exons, and acts as the host gene for miR-31, with miR-31 sequence residing within its intron 2. MIR31HG transcripts do not code for proteins but play a regulatory role in cellular processes such as proliferation, apoptosis, cell cycle regulation, EMT, senescence, and inflammation, largely through interaction with chromatin-modifying complexes, acting as a co-activator of hypoxia-inducible factor signaling, and modulating gene expression by sponging microRNAs and interacting with key transcription factors such as p300, HIF-1α, and MYC. Its dysregulation is implicated in a variety of cancers (often correlating with poor prognosis and tumor progression) and non-cancer pathologies, suggesting its promise both as a therapeutic target and a biomarker for diagnosis and prognosis. Therapy targeting MIR31HG or its epigenetic modulation is under exploration, but clinical application remains in early stages due to complexities of lncRNA targeting and functional diversity in different disease contexts.
