Overview

MIR365BHG (MIR365B and MIR4725 host gene) is a long non-coding RNA located at chromosome 17q11.2. It serves as the host gene for microRNAs miR-365b and miR-4725, which are transcribed and processed to regulate multiple downstream target genes via the RNA interference pathway. While MIR365BHG itself does not encode protein or serve as a classic drug target, its microRNA products function as critical regulators of gene expression in cancer progression, with reported roles in cell proliferation, apoptosis, invasion, and metastasis. Downregulation of miR-365 (a product of MIR365BHG) is associated with poorer prognosis in melanoma and non-small cell lung cancer, suggesting utility in biomarker development and possibly as a future avenue for RNA-based therapeutics. MIR365BHG's association with disease is indirect, through the biological activity of its microRNA products. Therapeutic approaches focusing on MIR365BHG would almost certainly involve modulation of its derived miRNAs rather than the host transcript itself.
