Overview

The **MIS12 kinetochore complex component (MIS12)** is a core protein subunit of the MIS12 complex, which is one of three main components of the KMN network—composed of KNL1, MIS12, and NDC80 complexes—forming the major microtubule-binding interface of the outer kinetochore. MIS12 plays a crucial structural role in chromosome segregation by linking centromeric chromatin (via direct interaction with CENP-C and CENP-T) to the force-generating microtubule attachment sites. It acts as a scaffold that coordinates and stabilizes assembly of the kinetochore, facilitating faithful segregation of chromosomes during mitosis. The MIS12 complex does not have enzymatic or receptor activity and is not directly targeted by drugs, but its disruption leads to severe mitotic defects and chromosomal instability, often associated with cancer and aneuploidy. The structure and function of MIS12 are highly conserved across eukaryotes, with critical regulatory contributions from mitotic kinases like Aurora B, which enhance MIS12 complex interactions through phosphorylation[1][2][3].
