Overview

Mis18-binding protein 1 (MIS18BP1) is a chromatin-associated protein essential for the assembly and maintenance of centromeres in human cells. It is a pivotal component of the Mis18 complex, functioning as a scaffold that recruits the Mis18α/β heterodimers to the centromere during the G1 phase of the cell cycle. MIS18BP1 is necessary for the accurate loading of CENP-A, a centromeric histone variant critical for kinetochore formation, thus ensuring faithful chromosome segregation during mitosis[2][7]. Loss or dysfunction of MIS18BP1 disrupts CENP-A incorporation, resulting in centromeric defects and chromosome mis-segregation. Its core molecular role is in centromere identity maintenance rather than classical signaling, enzymatic activity, or transport. Although vital for genome stability, MIS18BP1 is not a therapeutic target for drugs and no direct drug interactions are documented[2][3][7].
