Overview

Mitochondrial and cellular lipid membranes are fundamental structural components of eukaryotic cells, primarily composed of phospholipid bilayers that facilitate compartmentalization and regulate the movement of solutes. The mitochondrial inner membrane is uniquely enriched with cardiolipin, a specialized phospholipid essential for the structural integrity of cristae and the optimal assembly of the electron transport chain complexes (Birk et al., 2013, PubMed: 23255577). Pathological alterations in these membranes, such as lipid peroxidation or loss of cardiolipin, are central to the progression of heart failure, ischemia-reperfusion injury, and neurodegenerative disorders (Szeto, 2014, PubMed: 24630934). Therapeutic strategies targeting these membranes include the use of mitochondria-targeted peptides like elamipretide, which binds to cardiolipin to stabilize the membrane and restore bioenergetic function (Chavez et al., 2018, PubMed: 30012718). Additionally, cellular membranes serve as critical targets for various antimicrobial and antifungal drugs that exploit specific lipid compositions to induce membrane permeability and cell death in pathogens (Humphries et al., 2011, PubMed: 21613281).
