Overview

The mitochondrial and endothelial nitric oxide synthase (NOS) system comprises enzymes responsible for the production of nitric oxide (NO), a critical signaling molecule. Endothelial NOS (eNOS or NOS3) is primarily located in the vascular endothelium, where it regulates vascular tone, inhibits platelet aggregation, and maintains cardiovascular homeostasis (UniProt P29474). Mitochondrial NOS (mtNOS), often considered a localized isoform of neuronal NOS (nNOS) or eNOS, resides within the inner mitochondrial membrane and modulates cellular respiration by interacting with the electron transport chain (PubMed: 9106651). Together, these components play a vital role in regulating oxygen consumption, ATP production, and oxidative stress. Dysregulation of these enzymes is linked to endothelial dysfunction, hypertension, and metabolic disorders (Circulation, 2011). Pharmacological modulation often involves increasing NO bioavailability or preventing NOS uncoupling, where the enzyme produces superoxide instead of NO (StatPearls: NBK507826).

Mechanism of action

NOS enzymes catalyze the production of nitric oxide (NO) from L-arginine; NO activates soluble guanylate cyclase to increase cGMP, causing vasodilation and regulating mitochondrial respiration via cytochrome c oxidase inhibition (StatPearls: NBK507826; PubMed: 9106651).
