Mitochondrial DNA (mtDNA) synthesis is the process by which mitochondria replicate their own genetic material, a small circular double-stranded DNA molecule distinct from nuclear DNA. This process is essential for mitochondrial function and cellular energy production. It involves key enzymes like POLG, TWINKLE, and mitochondrial SSB protein, all encoded by nuclear genes and imported into mitochondria. Replication initiates at specific origins (OH and OL) using RNA primers synthesized by mitochondrial RNA polymerase.
