Mitochondrial import inner membrane translocase subunit Tim17-B (TIMM17B) is a core component of the TIM23 complex in the inner mitochondrial membrane, essential for translocation of proteins from the cytosol into the mitochondrial matrix or for sorting them into the inner membrane[1][2][3][5]. TIMM17B, together with other subunits such as Tim23 and Tim21, forms a dynamic structure involved in recognizing protein presequences and facilitating their import in a membrane potential-dependent fashion[1][3]. TIMM17B contains four transmembrane domains forming a lateral cavity crucial for presequence protein translocation, with conserved negative charges near the intermembrane space that mediate these interactions[1]. While not a classic drug target, TIMM17B's function is fundamental for mitochondrial health and overall cellular metabolism[1][2][3].
