Overview

Mitochondrial import inner membrane translocase subunit TIM50 (TIMM50) is a core component of the TIM23 translocase complex, essential for protein import across the mitochondrial inner membrane in eukaryotic cells. Acting as the receptor subunit on the matrix side, TIM50 recognizes targeting presequences of mitochondrial precursor proteins, facilitating their transfer from the TOM complex in the outer membrane through to the matrix or inner membrane. TIMM50's activity is critical for mitochondrial function, bioenergetics, and programmed cell death (apoptosis) pathways. Pathogenic variants in TIMM50 cause a spectrum of mitochondrial disorders, including epilepsy, intellectual disability, and 3-methylglutaconic aciduria. Dysregulation of TIMM50 expression has been linked to cancer proliferation and cardiac hypertrophy.

Mechanism of action

Antiepileptic drugs act on neural excitability to control seizures but do not directly target TIMM50; treatments address symptoms from TIMM50 dysfunction, not the protein itself. No known direct pharmacological inhibitors or activators of TIMM50 are established.
