Overview

The mitochondrial import receptor subunit TOM40 homolog (TOMM40) is a channel-forming protein located in the outer mitochondrial membrane and serves as the essential component of the translocase of the outer membrane (TOM) complex[1][3][8]. TOMM40 forms the pore through which most mitochondrial precursor proteins are translocated from the cytosol into the mitochondrion[1][2]. It plays a crucial role in mitochondrial protein import, homeostasis, and indirectly supports the function and assembly of the mitochondrial respiratory chain[3][8]. Genetic variants in TOMM40 are associated with increased risk for late-onset Alzheimer’s disease, potentially due to linkage disequilibrium with the APOE gene, and may have additional links to other neurodegenerative disorders and cardiovascular diseases[1][3][5]. There are currently no specific drugs approved that directly target TOMM40, but its variants may be acknowledged as biomarkers for Alzheimer’s disease risk stratification[5].
