Overview

Mitochondrial import receptor subunit TOM40B (TOMM40L) is a component of the translocase of the outer mitochondrial membrane (TOM) complex, which is essential for the import of the vast majority of nuclear-encoded proteins into mitochondria[5][1]. It is predicted to function as a channel-forming protein (pore-forming) that mediates translocation of cytosolic precursor proteins into the mitochondrial intermembrane space, serving as part of the protein import machinery necessary for mitochondrial biogenesis and function[5][6][3]. TOMM40L belongs to the Tom40 family and shares homology with the main TOMM40 (TOM40) but appears to be a less well-studied paralog with potential redundancy or specialized roles[5][3]. It is expressed in human tissues and localized to the mitochondrial outer membrane[6][5]. There are no reported direct small molecule drugs or established roles as a primary therapeutic target, but genetic variation in TOMM40L has been associated in some studies with risk modulation for neurodegenerative diseases such as Alzheimer's disease[4][5]. No direct mechanism-based drugs, biomarkers, or notable safety issues are currently established for TOMM40L.
