Overview

Mitochondrial metabolic machinery refers to the integrated system of enzymes, transporters, and protein complexes within the mitochondria that drive cellular energy production and metabolic homeostasis (NIH, 2024; Frontiers, 2021). This machinery includes the electron transport chain (ETC) for oxidative phosphorylation, the tricarboxylic acid (TCA) cycle, and pathways for fatty acid oxidation and amino acid metabolism (NIH, 2024; MDPI, 2024). In many diseases, such as cancer and type 2 diabetes, this machinery is rewired to support rapid proliferation or compensate for nutrient excess, leading to the production of oncometabolites or reactive oxygen species (ResearchGate, 2025; MDPI, 2024). Therapeutic strategies often target specific components of this machinery, such as Complex I or III, to induce metabolic stress or trigger apoptosis in diseased cells (NIH, 2024; MDPI, 2024). Furthermore, the machinery plays a critical role in regulating cell death pathways and calcium signaling, making it a central hub for cellular fate decisions (NIH, 2024). However, targeting these pathways requires careful management of safety concerns like lactic acidosis and systemic toxicity due to the essential role of mitochondria in all aerobic tissues (NIH, 2024; UGA, 2010).
