Mitochondrial nucleoid-associated protein 1 (MTNAP1) is a nuclear-encoded, mitochondrial membrane-associated protein predicted to play a critical role in controlling mitochondrial DNA copy number and replication. It binds double-stranded DNA throughout the mitochondrial genome without sequence specificity and promotes mtDNA replication, thereby influencing mitochondrial metabolism and cell proliferation. MTNAP1 is associated with the mitochondrial nucleoid and does not act as a receptor, enzyme, or transporter. Pathogenic variants in MTNAP1 have genetic associations with postpoliomyelitis syndrome and developmental and epileptic encephalopathy 28 in humans. Its wider biological and pathological significance, as implied by some aliases, is not confirmed by credible scientific databases. There are no drugs, mechanisms of action, biomarkers, or recognized safety considerations documented for MTNAP1.
