Overview

Mitochondrial outer membrane import complex protein 2 (MTX2), also known as Metaxin-2, is an outer mitochondrial membrane protein that forms part of the machinery responsible for the import of proteins into mitochondria. MTX2 interacts directly with other members of the metaxin family (MTX1, MTX3), structural proteins of the MICOS complex (such as IMMT/mitofilin, CHCHD3, CHCHD6), as well as components of the translocase of the outer mitochondrial membrane (TOMM6), and molecular motors involved in mitochondrial transport (MIRO-1, KLC-1, TRAK-1). MTX2 has a crucial role in maintaining mitochondrial integrity, biogenesis, morphology, and cellular energy metabolism. Genetic loss or mutation of MTX2 is linked to severe mitochondrial dysfunction, premature aging syndromes, and kidney disease. Increased expression of MTX2 in lung adenocarcinoma (LUAD) correlates with tumor growth, altered metabolism, and poorer patient survival, identifying MTX2 as a promising therapeutic target and prognostic biomarker in cancer. In neurons, MTX2 is essential for the proper transport of mitochondria into dendrites and axons, underlying its importance in neurodevelopment and neurodegeneration.
